Resumo

The Amazon biome contains a high and complex biodiversity, among the thousands of unexplored organisms, the endophytic fungus stands out for its ability to produce an arsenal of substances and enzymes of biotechnological interest. Therefore, the objective of this work was to carry out biotransformation reactions of compounds derived from keto-Imidazole, mediated by cells of endophytic fungi isolated from Bertholletia excelsa (Brazil Nut), so far not described in the literature, for the production of alcohols enantiomerically pure. The synthesis of ketoimidazole was performed, and then 5 strains of different endophytic fungi isolated from the almond of B. excelsa were cultivated in a 250 mL flask containing 100 mL of liquid medium (2% malt), previously sterilized. After this period, the mycelium was filtered and resuspended in a buffer solution (pH 7) with the addition of 25 mg of the compound solubilized in 400 μL of DMSO, remaining under controlled conditions (30 ºC, 120 rpm) for 7 days. Reactions were filtered and extracted with ethyl acetate, dried with sodium sulfate, and filtered again. The identification of the formed product, and conversion percentage for each reaction was performed by Gas Chromatography - Mass Spectrometry (GC-MS). In all cases the alcoholimidazole was identified as a biotransformation product, the product conversion rate ranged from >99 – 15%. The fungus Penicillium paxilli (BIORG 6) had a higher conversion rate (>99%), compared to the reaction with the fungus Aspergillus sp. (BIORG 9) promoted a lower conversion rate (15%). Other compounds derived from keto-imidazole were synthesized and used in biotransformation with the fungus Penicillium paxilli (BIORG 6)
